Yes, well sort of. You can use the Windows file encryption system to protect
the file or folder. Please
refer to Microsoft KB article.

Instead use NTFS Permissions:

Another suggested encryption.
Be very careful if you use EFS.
Improper use can and has lead to permanent loss of data:
