My boss is using XP Pro SP2 and up to a couple months ago was able to access
his machine from home using a VPN connection and the Remote Desktop program.

He has UltraVNC installed and used to have another NIC installed. It has
recently been uninstalled and the problem still remains.

Any ideas, gang?

Do I try this from a remote computer or from the affected machine?

That is, I get into hyperterminal, open a tcp/ip connection to his IP
address and use port 3389?

Is there a way of testing what is using port 3389?
